Yönlendirilmiş doku rejenerasyonu (YDR), yıllardır periodontal rejenerasyon için altın standart olarak kabul edilmekte ve kronik periodontitis hastaların klinik parametrelerinin iyileştirilmesinde etkili olduğu bilimsel literatürde ifadeedilmektedir.YDR'nin periodontoloji klinik uygulamaları; kemik ve dokunun yeniden büyümesini sağlamak ve kaybedilen periodontal yapıları yeniden oluşturmak için fiziksel bir bariyer sağlayacak olan membranın yerleştirilmesini kapsar. Bu çalışmada; cerrahi işlem sonrası oluşabilecek enflamasyonu engelleyecek ve lokal antienflamatuar ilaç salımını sağlayacak kitosan kaplı PCL membranların elektroeğirme yöntemi ile üretilmesi ve bu membranların periodontal yara iyileştirici potansiyellerin in vitro olarak araştırılması amaçlanmıştır.YDR amaçlı PCL membranların elektroeğirme yöntemi ile üretilmesi için optimizasyon çalışması yapılmış ve %20PCL (a/h) optimum polimer konsantrasyonu olarak tercih edilmiştir. Optimize edilmiş olan %20 PCL (a/h) fiberlere %5 ve %15 (a/h) Metronidazol yüklenmiş ve fiberler daha sonra kitosan %5 (a/h) ile kaplanmıştır. Üretilmiş olan membranların karakterizasyonu için viskozite analizi, optikmikroskop,FTIR, SEM,temas açısı analizi, mekanik, degredasyonve in vitro salım testi yapılmıştır. Optik mikroskop,FTIR ve SEM analizi sonuçları PCL fiberlerin başarılı bir şekilde oluşturulduğunugöstermiştir. FTIR ve SEM analizi sonuçları ile Metronidazolün PCL fiberlere başarılı bir şekilde yüklendiğini ve kitosan ile kaplandığı görülmüştür. SEM sonuçları karşılaştırıldığında Metronidazol miktarının %0'dan %15'e yükseltilmesi ortalama çapı 2,0±0,29µm'den 0,494±0,22 µm'ye, PCL'in kitosan ile kaplanması sonucu ise ortalama çapı 0,792±0,26µm'ye düşürmüştür. PCL'in temas açısı 114,51±2,10° iken, kitosan kaplı PCL'in 71,29±0,29°, %5 ve %15(a/h)Metronidazol yüklü PCL'in ise 76,02±4,46° ve 52,55±1,69°olarak hesaplanmıştır. Temas açısı sonuçları, PCL'in kitosan ile kaplanması veMetronidazol yüklenmesinin, PCL'in hidrofilik özelliğini arttırdığını göstermektedir. Mekanik test analizi sonuçlarına göre PCL'in young modülü 25,56±3,99 MPa ve çekme gerilim direnci 3,64±0,53 mm/mm iken, kitosan eklenmesiyle mekanik direnci zayıflamış ve young modülü 5,04±1,16 MPa ve çekme gerilim direnci 1,67±0,37mm/mm olarak ölçülmüştür.Yirmi sekiz gün boyunca yapılan degredasyon çalışması sonuçları, kitosan kaplı PCL'in 28. günün sonunda % 93'ünün bozunduğunu ve bu süreç sonunda fiber yapısını hala koruyabildiğini göstermiştir. Guided tissue regeneration (GTR) has been accepted as the gold standard for periodontal regeneration for many years and has been reported to be effective in improving the clinical parameters of patients with chronic periodontitis. Clinical applications of periodontology of GTR; encompassing the placement of a membrane that will provide a physical barrier to regenerate bone and tissue and to regenerate lost periodontal structures. In this study; The aim of this study was to produce chitosan coated PCL membranes by electrospinning method to prevent inflammation and local anti-inflammatory drug release and to investigate periodontal wound healing potential of these membranes in vitro. An optimization study was carried out to produce PCL membranes for GTR by electrospinning method and 20% PCL (w/v) was chosen as the optimum polymer concentration. The optimized 20% PCL (w/v) fibers were loaded with 5% and 15% (w/v) Metronidazole and the fibers were then coated with chitosan 5% (w/v). For the characterization of produced membranes, viscosity analysis, optical microscope, FTIR, SEM, contact angle analysis, mechanical, degradation and in vitro release tests were performed. Optical microscope, FTIR and SEM analysis results showed that PCL fibers were successfully formed. Results of FTIR and SEM analysis showed that Metronidazole was successfully loaded into PCL fibers and coated with chitosan. When the SEM results were compared, increasing the amount of Metronidazole from 0% to 15% mean diameter was 2.0±0.29 µm to 0.494±0.22 µm, and PCL was coated with chitosan with a mean diameter of 0.792±0.26 µm. PCL had a contact angle of 114.51±2.10°, while 71.29±0.29° 5%, and 15% (w/v) of PCL coated PCL had 76.02±4 46° and 52.55±1.69°. Contact angle results show that PCL coating with chitosan and Metronidazole loading increase the hydrophilicity of PCL. According to the results of the mechanical test analysis, the young modulus of PCL was 25.56±3.99 MPa and the tensile strength was 3.64±0.53 mm/mm. and tensile stress resistance was calculated as 1.67±0.37mm/mm. The results of the 28-day degredation study showed that 93% of chitosan-coated PCL decayed at the end of 28th day and still retain the fiber structure at the end of this process. In vitro release studies of 5% and 15% (w/v) Metronidazole from membranes were performed at 25° C and 37° C for 28 th day. At the end of the 28th day, drug release of metronidazole at 25° C was 72.04±0.09%, while drug release at 37° C was 91.44±0.37%. After the analyzes, the potential of the Metronidazole loaded membranes to be used in the biomedical and tissue engineering applications has been successfully demonstrated.

Periodontitis, periodontal dokularınzarar görmesine neden olan ve diş kaybı ile sonuçlanan kronik inflamatuar bir hastalıktır.
